Flex Automation

High-throughput system capable of handling up to 20x 384-well plates in parallel for temperature or enzamatically controlled reactions. Currently set up for Golden Gate Assemblies.

Immunofluorescence Staining Protocol

OT2

Using an Opentrons-OT2 to perform the following protocol.

  • Remove excess liquid
  • PBS wash 3x
  • Add & remove permeabilization solution
  • Add & remove blocking solution
  • Add primary antibodies and incubate overnight
  • PBS wash 3x
  • Add secondary antibodies solution
  • BPS wash 3x
  • Add & remove DAPI
  • PBS wash 3x
  • Add PBS + Sodium Azide

Tecan

Periodic measurements every 30 minutes over a 4 day period

  • Incubate 96-well plates at 37 °C
  • Monitor growth via absorbance at 600 nm
  • Measure fluorescence
  • De-lid plate prior to measurement
  • Run multiple plates in parallel
