Within the external page NCCR Molecular Systems Engineering I'm investigating design rules for synthetic operons. While progresses in synthetic biology are leading to more and more efficient answers to the question “How to write DNA?”, the question of “What to write into the DNA?” remains challenging. I aim to answer this question in regard to bacterial polycistronic operon systems. Such operons, in which multiple genes are expressed from only one promoter, are an important functional unit in bacteria. In order to gain enough knowledge to engineer future multi-protein systems I am deploying a top-down approach, where high-throughput DNA assembly helps me to construct large numbers of variants and multiplexed multilevel assays provide me quantitative information about the system parameters or DNA copy-numbers, mRNA amounts, and protein level and activity.

Furthermore, I'm interested in developing bioinformatic tools that allow efficient screening for fine tuned protein expression levels in synthetic systems, e.g. the algorithm for generating smart RBS libraries:
